ISP 4

International Streptomyces Project medium 4, also named inorganic salts-starch agar

Preparation

  • 10 g Soluble Starch
  • 1 g MgSO4 x 7H2O
  • 1 g NaCl
  • 2 g (NH4)2SO4
  • 2 g CaCO3
  • 1 L dH2
  • 1 mL Trace Salts solution
    • 0.1 g FeSO4 x 7H2O
    • 0.1 g MnCl2 x 4H2O
    • 0.1 g ZnSO4 x 7H2O
    • 100 mL dH2O
  • 20 g agar


Instructions

Prepare as two separate solutions and mix before adding agar and autoclaving

Solution 1

  1. Make a paste of the soluble starch with a small volume of cold dH2O
  2. Bring the paste up to a volume of 500 mL

Solution 2

  1. Dissolve the remaining constituents and trace salts solution in 500 mL dH2O
  2. Adjust the pH so that it's between 7 and 7.4

Uses

  • Screening for antibiotic production
