pCAP03
The pCAP03 plasmid has been successfully used to create capture vectors that can be used for TAR cloning of biosynthetic gene clusters.
Background
The plasmid was developed by Bradley Moore's lab as an upgraded version of pCAP01.
Construction
Uses
Characteristics
Capture arms can be cloned within the restriction enzymes sites, and the plasmid can be selected for with kanamycin. pCAP03 contains a URA3 gene that allows for selection against non-homologous end joining in the presence of 5-fluoroorotic acid (5-FOA). pCAP03 also contains an apramycin resistance cassette (acc(3)IV).
Map and Sequence
The plasmid map can be viewed on AddGene here, where the plasmid can also be purchased.
